3、populationofthispathogeninthisprovinceInthepresentstudy“dahliaew那isolatedfromdiseasedstemsofcottoncollectedfromHubeiProvince,TheseisolatestogetherwithtwoLDisolates①andV991)wereusedto:(1)compareculturalcharacteristicsonsu

4、bstratesincludingpotatodexaoseagar(PDA)andautoclavedcottonseedshells(ACS);(2)testpathogenicitydifferentiationonfourcottonvarieties;and(3)disclosethegeneticdiversityusingrandomamplifiedpolymorphicDNA(RAPD)assaysFrommainco

5、ttonproducingareasinHubeiProvince,15isolatesofIfdahliaewereobtainedandsingleconidium(SC)isolationwasmadetofurtherpIlri母theseparentalisolatesCulturalstudiesshowedthatgrowthrate,sporeproductionandcolonymorphologyvariedamon

6、gtheseSCisolatesThegrowthraterangedbetween16and23cm/dandsporeproductionchangedfrom069x108to2OOxl0S/gfreshACSsubslrateHowevertherelevantCO—efficientbetweenthegrowthrateandsporeproduction(r00781WaSnotsignificantlydifferent

7、(a=005)AfterthecultureswereincubatedOilPDAfbrl5days,threeculturaltypes,white,blackandgray(intermediate),wereobserved,andl,11and5isolateswereincludedineachgroup,respectivelyAfterthecultureswereincubatedonACSfor30days,anot

8、herthineculturaltypes,snowwhite,milkywhiteandgraycouldbeclaSsifiedand1,4and12isolateswereincludedineachgroup,respectivelyThepathogenicitydifferencebetweentheisolatesWaSdetectedonthefourcottonvarietiesOneachcottonvarietyH

9、ubeiisolatesshowingnodifferenceinpathogenicityfromT9orV991we∞observed111eresultsofthepathogenicitytestsalsoshowedthatZhongmian12wagmotetolerantthantheotherthreecottonvarietiestoIfdahliaeRAPDassaysshowedthat10outof27rando

10、mprimers(10mer)couldamplifythetemplateDNA(genomic)extractedfromthe17isolatesof14dahliaeThesimilaritycoefficientbetweentheisolatescalculated011thebasisoftheamplifiedDNAfragmentsWas037100ClusteranalysisrevealedthattwAPDgro
